Cell Based Kinase/Phosphorylation Assays

  • Cellular Phosphorylation Assay

This assay quantifies the phosphorylation status of a substrate in cells. It can be performed with cells  expressing either endogenous or overexpressed kinases.

The phosphorylation is detected using phospho-site-specific antibodies via ELISA or AlphaLISA.

  • Cell-Deliverable Peptide Substrate Assay

Uses cell-penetrating peptides designed with kinase substrate sequences. These peptides are taken up by cells and phosphorylated by active kinases. Phosphorylation is then measured using ELISA or other detection methods.

 

Cell Based Reactive Oxygen Species (ROS) Assays

  • Fluorescence-Based Assays

H₂DCF-DA (2′,7′-Dichlorodihydrofluorescein Diacetate) and DHE (Dihydroethidium) assays to detect superoxide radicals in cells.

  • CellROX Reagents

Used for detecting ROS in live cells using imaging, flow cytometry, or microplate analysis.

  • MitoSOX Red

Used to detect superoxide production in mitochondria.

  • CM-HDCFDA (Chloromethyl-HDCFDA)

Used for detecting ROS in live cells, particularly hydrogen peroxide, superoxide, and hydroxyl radicals.

  • Chemiluminescence-Based Assays

Used for detecting various ROS, including hydrogen peroxide, superoxide, and hydroxyl radicals in biological samples.

  • Spectrophotometric Assays

FOX (Ferric-Xylenol Orange) Assay

  • Genetically Encoded Sensors

roGFP (Redox-Sensitive Green Fluorescent Protein)

 

Cell Based Nitric Oxide Synthase (NOS) Assays

  • Intracellular Nitric Oxide Assay (Fluorometric)

 Utilizes a nitric oxide-specific fluorescent probe that is highly photostable and has a wide pH stability range.

  • Homogeneous Fluorescent Cell-Based Assay

It involves the use of ionomycin to stimulate NO production, which is then detected using a fluorescent probe.

  • Nitric Oxide (NO) Assay Kit (Colorimetric)

Measures total NO by quantifying nitrite and nitrate, the stable end products of NO oxidation.

  • In Vitro Nitric Oxide (NO) Assays (Colorimetric and Fluorometric)

These assays measure NO indirectly by detecting its breakdown products (nitrite and nitrate) in various biological samples.
