T Cell Assays

Mixed Lymphocyte Reaction is a type of T cell proliferation assay.  

  • T cell activation assays

Activation of T cells through the T Cell Receptor (TCR) results in cellular changes.

CD69, CD25 up-regulation by flow cytometry.

ELISAs for cytokine / chemokine expression.

Phospho-specific western blots to evaluate kinase / phosphatase activity.

  • Cytotoxicity assays

A T cell cytotoxicity assay measures the ability of T cells to kill target cells, indicating immune response efficacy.Cytotoxic T cells, also known as CD8+ T cells, are responsible for killing target cells.

  • T-cell exhaustion assay

Infection and cancer can lead to cellular exhaustion. Inability of T cells to respond to stimuli is evaluated. Cytokine response assays, Cytotoxicity assays, Flow cytometry assays.

  • T Cell Suppression assay

The effect of immunomodulatory agents on T cell function is measured, particularly in the presence of regulatory T cells (Tregs).

  • Immune Checkpoint Assays

The expression levels of inhibitory checkpoint molecules (e.g., PD-1, CTLA-4) on T cells is measured using flow cytometry, ELISA, and reporter assays.

  • Memory T cell recall assays

Memory T cells can be generated in humans by a number of antigenic stimuli including tumor antigens,

infection and vaccines. Recall of memory T cells in human PBMC are measured by ELISPOT and flow cytometry.

Different T cell populations are identified and quantified using antibodies against specific surface markers.  

  • Intracellular Cytokine Staining (ICS)

The production of cytokines within T cells after stimulation is measured.

  • T cell proliferation assays

 This assay measures the proliferation of T cells in response to stimuli such as antigens or mitogens.

Macrophage Assays

  • Cytokine Secretion Assays

The production of cytokines by macrophages is measured using ELISA or multiplex bead-based flow cytometry assays.

  • Surface Marker Expression Assay

Flow cytometry is employed to analyze the expression of surface markers indicative of macrophage activation and polarization (e.g., CD80, CD86 for M1; CD206, CD163 for M2).

  • Phagocytosis Assay

Macrophages are effective at killing tumor cells through phagocytosis after recognition of cellular damage or binding to antibodies (antibody mediated cellular phagocytosis, ADCP) which can be measured in vitro by flow cytometry

  • Polarization Assay

These assays differentiate macrophages into M1 (pro-inflammatory) or M2 (anti-inflammatory) phenotypes using specific cytokines like IFN-γ and IL-4 and it is quantified using flow cytometry.

Neutrophil Assays

  • Oxidative Burst Assay

This assay measures the production of reactive oxygen species (ROS) by neutrophils in response to stimuli.

  • CD62L Shedding Assay

The shedding of CD62L (L-selectin) from the surface of neutrophils into the supernatant media is measured as an indicator of activation.

  • Degranulation Assay

The release of granule contents from neutrophils, such as myeloperoxidase or elastase, both components of Neutrophil Extracellular Traps (NETs) are measure using custom ELISAs.

  • Chemotaxis assay

The chemotactic ability of cells, which is their directed movement in response to chemical stimuli is evaluated by microscopy.

Dendritic Cell (DC) Assays

  • Inflammasome Activity Assay

Inflammasome assays are used to evaluate the activation of inflammasomes within dendritic cells.

  • Mixed Lymphocyte Reaction (MLR) Assay

Co-cultures of PBMCs or DCs with allogeneic T cells and/or drugs are evaluated for proliferation to ascertain activation.

  • Cytokine Production Assay

PBMCs synthesize and secrete specific cytokine by PBMCs or DCs in response to various stimuli which are measured by FLOW or ELISA.

Natural Killer (NK) Cell Assays

  • Tumor cell killing

Human PBMC derived NK cells are cultured with tumor cell lines and evaluated for killing activity by flow cytometry.

Tumor targeting antibodies enhance killing by NK cells .

  • Cytokine secretion by ELISA

Activated NK cells produce high levels of cytokines such as IFN-g.

NK cells up and down regulate a number of cell surface receptors such as 4-1BB after activation.

B-Cell Assays

Flow cytometry is used to analyze the expression of surface and intracellular markers on B cells.

  • ELISA (Enzyme-Linked Immunosorbent Assay)

ELISA is used to quantify the amount of specific antibodies produced and secreted by B cells in response to an antigen.

  • B-cell Proliferation Assay

This assay measures the proliferation of B cells in response to stimuli such as antigens or mitogens.

  • B-cell Activation Assay

This assay assesses the activation status of B cells by measuring the expression of markers (e.g., CD69, CD86) and cytokine production.

  • B-cell Cytokine Production and Secretion Assay

This assay measures the production of cytokines by B cells in response to stimulation.

  • B-cell Differentiation Assay

This assay studies the differentiation of B cells into plasma cells or memory B cells.
