Flow Cytometry

Immunobiology, drug discovery and flow cytometry

Flow cytometry is a technology that identifies individual cells labeled with a dye or marker by passing the cell through a laser beam. Flow is used to evaluate therapies that involve regulating immune responses.  Some disease indications where the immune system can be therapeutically modulated include: Autoimmunity / Inflammation, Cancer and Infectious disease. Flow cytometry is commonly used to evaluate different drug modalities:

  • Small molecule inhibitors of extracellular and intracellular immune responses
  • Antibody Drug Conjugates (ADC)
  • Immune modulating antibody-based therapies: bi and tri-specific, T cell engagers, immuno-cytokines etc.
  • Cellular Therapies: CAR T cells, other genetically modified T cells, tumor-infiltrating lymphocytes (TIL), macrophages, and NK cells
  • Protein modifiers

We maintain direct communication with the FDA, both independently and through our clients, ensuring compliance and smooth regulatory processes.

Our Commitment to Excellence

MarinBio’s scientists have perfected the art of flow cytometry, creating assays for the past 30 years that can analyze cells in complex matrices such as blood, plasma, serum, and cell culture media. We’ve developed reliable and sensitive Flow Cytometry protocols for our clients, customized to meet specific pre-clinical and clinical needs.

We maintain direct communication with the FDA, both independently and through our clients, ensuring compliance and smooth regulatory processes.

MarinBio has successfully developed Flow Cytometry for the following applications, among others:

  • Cell phenotyping, cell differentiation, T-lymphocyte subset analyses
  • Ligand-binding, -inhibition and -specificity of binding
  • Cell-activation, -growth inhibition, -proliferation,
  • MLR (Mixed Lymphocyte Reaction)
  • Apoptosis
  • Gene expression
  • Protein expression
  • Drug-induced-signaling pathways, cytokine production
  • Multiplexquantitation of different analytes in a single sample
  • Protein post-translational modifications, including cleaved and phosphorylated proteins to study

intracellular signaling pathways

  • Cell cycle analyses, providing a powerful tool to assess cells in G0/G1 phase versus S phase,

G2 phase or polyploidy and including analyses of cell proliferation and cell activation

  • Cell health and metabolic status, ranging from cell viability to late stage programmed cell death

Other Capabilities:

  • Isolation of single cells with desired phenotypic characteristics by a process called cell sorting.
  • Cell-based applications include analyzing cell metabolism, cell-health, cell viability, small molecules, nucleic acids (DNA and RNA), proteins, or chromosomes

Case Study:

Flow cytometry analysis of lung leukocyte population:

Digested lung samples are analyzed using two parallel antibody panels to study Myeloid

and Lymphoid cell populations.

Top panel: Myeloid cells where selected by Forward and Side Scatter properties and live single cells where further phenotyped using cell-specific antibodies to obtain

NeutrophilMacrophageMonocyte and  Alveolar Macrophage counts.

Bottom panel: Lymphoid cells where selected by Forward and Side Scatter properties and live single cells where further phenotyped using cell-specific

antibodies to obtain T-cell (CD4 or CD8 positive), B-cell and NK-cell (Natural Killer) counts.
