Case Study

Flow cytometry analysis of lung leukocyte population

Digested lung samples are analyzed using two parallel antibody panels to study Myeloid and Lymphoid cell populations.

Top panel: Myeloid cells where selected by Forward and Side Scatter properties and live single cells where further phenotyped using cell-specific antibodies to obtain Neutrophil, Macrophage, Monocyte and  Alveolar Macrophage counts.

Bottom panel: Lymphoid cells where selected by Forward and Side Scatter properties and live single cells where further phenotyped using cell-specific antibodies to obtain T-cell (CD4 or CD8 positive), B-cell and NK-cell (Natural Killer) counts.

Immunobiology, drug discovery and flow cytometry

Flow cytometry is a technology that identifies individual cells labeled with a dye or marker by passing the cell through a laser beam. Flow is used to evaluate therapies that involve regulating immune responses.  Some disease indications where the immune system can be therapeutically modulated include: Autoimmunity / Inflammation, Cancer and Infectious disease. Flow cytometry is commonly used to evaluate different drug modalities:

  • Small molecule inhibitors of extracellular and intracellular immune responses
  • Antibody Drug Conjugates (ADC)
  • Immune modulating antibody-based therapies: bi and tri-specific, T cell engagers, immuno-cytokines etc.
  • Cellular Therapies: CAR T cells, other genetically modified T cells, tumor-infiltrating lymphocytes (TIL), macrophages, and NK cells
  • Protein modifiers
